I recently unlocked my 2nd hand iphone 3gs, but it is registered to the previous owners phone, if I factory reset the phone would I have to unlock it again?

No. It is activation locked, and only the person who set the lock can remove it.

I am afraid Lawrence may have misunderstood the question. You have an iPhone 3GS running iOS 6.1.6 so Activation Lock does not apply. I believe that by unlocking, you are referring to releasing a carrier lock. In that case, once it is unlocked, restoring the phone will not require unlocking again.
